Hepatotoxicity Assessments with Human Liver Spheroids

CYP3A4 Activity
CYP3A4 is the main hepatic cytochrome P450 expressed postnatally, making up about 30% of the liver's total P450 content. It plays a crucial role in drug metabolism. The levels of detectable CYP3A4 directly reflect the liver's detoxification capacity. A significant reduction in CYP3A4 levels indicates a serious impairment of liver function.

AST/ALT Activities
Drug-induced liver injury (DILI) is a well-known condition that can present symptoms similar to both acute and chronic liver diseases. Clinically, DILI can be categorized as hepatocellular, cholestatic, or mixed, depending on liver enzyme results. Elevated levels of ALT and AST are the primary indicators of DILI, reflecting the extent of liver damage. Consequently, it is essential to monitor changes in AST and ALT levels.

Hepatotoxicity
The toxicity of a drug to cells is closely linked to changes in cell number. As a result, the number of live cells serves as a direct indicator of the extent of growth inhibition caused by the tested agents.

Whole Blood Activation & Cytokine Release
Human whole blood was stimulated with different concentrations of Dynabeads (left) or PHA (right), and the levels of IL-2 and IFN-γ were measured using ELISA. The results show that the secretion of both IL-2 and IFN-γ is dose-dependent in response to Dynabeads or PHA stimulation.
